Studies of fibronectin synthesized by cultured chick hepatocytes   总被引:3,自引:0,他引:3  
We have adapted a chick embryo liver cell system for studying the synthesis of proteins secreted by hepatocytes. In primary liver cell cultures maintained for several days in arginine-deficient medium containing ornithine (0.7 mM) and carbamyl phosphate (1 mM), only hepatocytes demonstrated normal morphological and biosynthetic characteristics, indicating that they possessed a functional ornithine cycle as a source of arginine production. Non-parenchymal liver cells, such as fibroblasts, which lack the ornithine cycle were excluded. Hepatocytes in arginine-deficient or arginine-containing medium synthesized fibronectin (Fn) over several days at a constant rate of 3 micrograms +/- 1 microgram/mg cell protein per day, with fibronectin representing approximately 3% of the total secreted hepatocyte proteins during any culture period after the first 24 h. Pulse-chase experiments indicated that Fn synthesis and secretion was relatively rapid (t1/2 = 45 min) and represented approximately 95% of the intracellularly labelled Fn. This Fn is secreted predominantly as a 450 kD dimer with a subunit size that is indistinguishable from the plasma form as assessed by one-dimensional electrophoretic analysis. Continuous exposure of hepatocytes to insulin caused a moderate decrease (26%) in Fn synthesis, whereas there was no effect of short-term exposure. In contrast, dexamethasone stimulated Fn production 2-3-fold, consistent with its known ability to stimulate hepatocyte production of acute phase proteins. Under these conditions, electrophoretic analyses showed that an increased quantity of intact hepatocyte Fn was produced having the same molecular size of plasma Fn.  相似文献

The volatile metabolites of three strains of Pseudomonas aeruginosa and one strain each of Pseudomonas cepacia, Pseudomonas maltophilia, Pseudomonas fluorescens, and Pseudomonas putida were analyzed using an automated headspace concentrator incorporating a gas chromatograph. The procedure does not require sample preparation and automates the entire analytical sequence to yield reproducible profiles of volatile constituents. Gas chromatographic profiles of the volatile metabolites of each species were obtained using a 20-min concentration period and two fused silica capillary columns of different polarities. The production of headspace metabolites from trypticase soy broth was studied in relationship to culture incubation time and initial cell concentration. The volatiles identified after 24 h incubation consisted of 1-butanol, isopentanol, toluene, 1-undecene, 2-butanone, 2-heptanone, 2-nonanone, and 2-undecanone. Sufficient amounts of specific metabolites were produced after 5 h incubation to provide information of possible diagnostic value. In particular, all P. aeruginosa strains produced a distinctive series of 1-undecene and methyl ketones after 5 h incubation of media inoculated to provide 2 X 10(6) cells/mL. The results indicate that when growth and analytical conditions are held constant, P. aeruginosa and related species produce characteristic profiles of headspace metabolites. Since conventional bacteriological tests require 24 h or more for the identification of these pseudomonads, automated volatile analysis could provide an alternative means for the rapid detection of these bacteria.  相似文献

Aminoglutethimide (AG: 750 mg/day) was administered to a patient with idiopathic hyperaldosteronism (IHA) and circadian rhythms in urinary excretion of sodium (UNaV), potassium (UKV), aldosterone (AER) and 17-OHCS were analyzed by the single cosinor method. Urine was collected every 4h for 24h on the day before and on the 1st, 3rd and 7th day of AG administration, and above variables in each sample were determined. Circadian rhythms of 14 patients with primary aldosteronism (PA) who served as controls were also analyzed. In the present case, circadian acrophases in UNaV and AER studied before AG administration occurred at 22(19) and 07(05), respectively. They were similar to those of preoperative PA-patients. Circadian acrophase in UNaV occurred earlier with AG administration and on the 7th day it was at 14(05), a value similar to that of postoperative PA-patients. Circadian mesor in AER decreased remarkably from 4.1 to 0.6 micrograms/4h with AG administration, as did circadian mesor in UKV, whereas circadian mesor and acrophase in 17-OHCS did not change. Thus, the circadian characteristics in urinary variables in the present IHA-case were pathophysiologically similar to those of PA.  相似文献

The anticonvulsive action of diazepam, carbamazepine, sodium valproate and their combinations with pyridoxal-5-phosphate, nicotinamide, and alpha-tocopherol were investigated in acute experiments on mice with corazole-induced seizures. Diazepam (0.5 mg/kg), carbamazepine (50 mg/kg) and sodium valproate (200 mg/kg) were shown to reduce convulsive intensity and lethality. Vitamins nicotinamide (250 mg/kg), pyridoxal-5-phosphate (10 mg/kg) and alpha-tocopherol (100 mg/kg) potentiated anticonvulsive action of the above antiepileptic drugs. The results of the investigation suggest the efficacy of pathogenetic therapy and give new evidence of the advisability of using vitamins in combination with synthetic anticonvulsive drugs.  相似文献

Immune function in higher vertebrates is mediated primarilyby multimeric glycoproteins found in the serum and on the surfacesof lymphoid cells. These molecules possess common structuralfeatures suggesting that they belong to a supergene family whichmay have originated from a common ancestral gene. Some multigenicmembers of the supergene family undergo unique forms of chromosomalrearrangement during somatic development. We have identifiedimmunoglobulin heavy chainvariable region (VH) homologs in speciesrepresenting critical points in the vertebrate radiation, examinedtheir nucleotide sequences and found high degrees of organizationalhomology as well as localized regions of extended nucleotide(and amino acid) sequence identity with mammalian VH genes.The unexpected high degree of nucleotide sequence identity suggeststhat within this multigene family, selection may be operatingat both the DNA and polypeptide levels.Using several differentapproaches, the VH gene families in lower vertebrates have beenshown to be remarkably complex, discounting the possibilitythat a reduced number of germline genes accounts for the apparentlyrestricted natureof lower vertebrate immune responses. The lowervertebrate germline VH genes possess prototypic recombinationsignal sequences, implicated in the somatic reorganization ofmammalian immunoglobulin variable region genes, and segmentalreorganizationresembling that seen in mammals has been observed in an elasmobranch.The detection of a recombination element flanked by short, directrepeats within the intervening sequence of one reptilian VHgene suggests that these sequences may be mobile, perhaps functioningoutside of the immunoglobulin loci in other developmental processes.The complex nature of the variable region gene families andtheir capacity to undergo structural change during somatic developmentsuggest that unique genetic mechanisms may govern their evolutionarystabilization and diversification.  相似文献

A combined immunological study of cell-mediated and humoral immunity factors in sportsmen at the period of important competitions was made. The effect of the maximum bearable load was found to lead to the formation of a great number of universal rosette-forming lymphocytes and neutrophils. In the control group their number did not exceed 1-4% of the total population of these cells. Simultaneously, the suppression of the phagocytic activity of neutrophils and some of the humoral factors of systemic and local immunity with the parallel development of compensating processes were recorded.  相似文献

We have used immunocytochemistry and molecular cloning methods to identify and characterize structural polypeptides of the centromere. These studies permit us to resolve two distinct regions: the inner and outer centromere. (i) Components of the outer centromere: autoantibodies from certain patients with rheumatic disease identify a family of three immunologically related polypeptides that we have designated CENP-A (17 kDa), CENP-B (80 kDa), and CENP-C (140 kDa). CENP-B has been cloned and sequenced. DNA sequence analysis indicates that this polypeptide possesses two large regions with extraordinary concentrations of acidic residues (region I: 61 residues with 79% glu + asp; region II: 31 residues with 87% glu + asp). Despite this concentration of negative charge, immunocytochemical experiments suggest that CENP-B may be a DNA binding protein. In these experiments, the levels of CENP-B are seen to vary reproducibly from chromosome to chromosome. The role of CENP-B in vivo is unknown. However, it is unlikely to bind directly to the spindle microtubules since it is found at an inactive centromere that apparently does not attach to the spindle. (ii) Components of the inner centromere: we have injected mice with the whole chromosome scaffold fraction to elicit production of monoclonal antibodies. One such antibody identifies two structurally related polypeptides (the INCENP antigens, 135 and 155 kDa) that are preferentially located between the sister chromatids at the centromere. The INCENP antigens undergo dramatic movements from the chromosomes to the central spindle during mitosis. They are ultimately sequestered in the midbody and discarded. Several lines of evidence suggest that the INCENP polypeptides may be involved in the regulation of sister chromatid separation at the metaphase-anaphase transition.  相似文献
